چکیده

Protease inhibitors play a role in regulating proteases during cellular development and in plant defense. We cloned and sequenced cDNA encoding six protease inhibitors expressed in soybean roots infected with soybean cyst nematode (SCN) and determined their expression patterns. Four of these protease inhibitors are novel and have not been reported previously. Using RT-PCR, we measured the relative transcript levels of each protease inhibitor in roots of the soybean cv. Peking inoculated with either SCN TN8 to examine the expression of protease inhibitors during the susceptible interaction or with SCN NL1-RHg representing the resistant interaction. Within 12 to 24 hours, mRNA transcripts encoding five of the six protease inhibitors were more highly elevated in soybean roots exhibiting the susceptible interaction than the resistant interaction. Transcripts encoding two protease inhibitors possessing Kunitz trypsin inhibitor domains were induced 37- and 27-fold in the susceptible interaction within 1 dpi, but were induced only 5- to 7-fold in roots displaying the resistant interaction. Our results indicate that soybean roots recognize differences between these two SCN populations before the nematodes initiate a feeding site, and accordingly the roots express transcripts encoding soybean protease inhibitors differentially. These transcripts were generally less abundant in roots exhibiting the resistant interaction.

Emerging approaches to broaden resistance of soybean to soybean cyst nematode as supported by gene expression studies.

The major pest of soybean (Glycine max) is the soybean cyst nematode (SCN), Heterodera glycines. One population of SCN can evoke a resistant response while a second population can evoke a susceptible response from the same soybean cultivar. Recently, interactions between SCN and soybean roots have been studied using commercially available microarrays to measure both soybean and nematode transcr...

Defense Response of Soybean (glycine Max) to Soybean Cyst Nematode (heteroderaglycines) Race 3 Infection

Soybean plant responses to Heterodera glycines (H. glycines) race 3 infection were compared between resistant (Peking, LiaoK89102 and Franklin) and susceptible cultivars (Liao dou 10 and Kaiyu 10) at 10, 17, 20, and 23 days after inoculation (DAI). After inoculation with H. glycines race 3, the isozymes of peroxidase, polyphenol oxidase, superoxide dismutase, and catalase from the soybean roots...

Analysis of expressed sequence tags from roots of resistant soybean infected by the soybean cyst nematode.

The soybean cyst nematode (SCN) Heterodera glycines is the most devastating pest of soybean in the U.S.A. The resistance response elicited by SCN in soybean is complex, and genes involved in the response to a large extent are unknown and not well characterized. We constructed cDNA libraries made from mRNA extracted from roots of the resistant soybean Glycine max L. Merr. 'Peking' at 12 h, 2 to ...

Novel MiRNA and PhasiRNA Biogenesis Networks in Soybean Roots from Two Sister Lines That Are Resistant and Susceptible to SCN Race 4

The soybean cyst nematode (SCN), Heterodera glycines, is the most devastating pathogen of soybean worldwide. SiRNAs (small interfere RNAs) have been proven to induce the silencing of cyst nematode genes. However, whether small RNAs from soybean root have evolved a similar mechanism against SCN is unknown. Two genetically related soybean sister lines (ZP03-5373 and ZP03-5413), which are resistan...

RNA-seq data comparisons of wild soybean genotypes in response to soybean cyst nematode (Heterodera glycines)

Soybean [Glycine max (L.) Merr.] is an important crop rich in vegetable protein and oil, and is a staple food for human and animals worldwide. However, soybean plants have been challenged by soybean cyst nematode (SCN, Heterodera glycines), one of the most damaging pests found in soybean fields. Applying SCN-resistant cultivars is the most efficient and environmentally friendly strategy to mana...
